Matrix tablets based on a novel poly (magnesium acrylate) hydrogel for the treatment of inflammatory bowel diseases

The objective of this work was to evaluate the potential use a new polymer (PAMgA) in development sustained release matrix tablets for treatment bowel inflammatory diseases. For purpose, budesonide, highly lipophilic compound, used as model drug. Tablets with two reticulation grades PAMgA (PAMgA 5 and 40) 9 mg budesonide were developed characterized. All studies carried out using biorelevant media (FaSSGF FaSSIF). Swelling erosion influenced by grade assayed, being water uptake higher 40 intestinal fluid, whereas showed more intense medium. Budesonide released slowly from tablets, both gastric environment, following Super case II transport kinetics (relaxation-controlled delivery), lag time around 1–2 h. When dissolution medium changed sequentially throughout trial, 75% dose manner between 4 20 h testing showing controlled than Entocort® Budenofalk® (commercially available formulations budesonide). In conclusion, allows controlling drugs an useful excipient tablets.
